    Castres 2013 - 2017 Skin aging and cumulative exposure to UVs within a lifetime affects a number of actors in the extracellular matrix such as collagen, elastin, glycans and associated biomolecules (metalloproteinases and their inhibitors, hyaluronidases, etc). The concentration, structure and / or distribution, and the hydric organization are modified by this phenomenon. All these modifications result in a loss of function of the matrix and the clinical manifestations of aging.
    The aim of this thesis, carried out in collaboration with the Skin Research Center of Pierre Fabre Dermo-Cosmétique, is to analyze the changes in the dermis and the hydric organization within the matrix with age and under the effect of UVs to help the knowledge of skin alteration under these conditions. Given the different scales to be explored - from Angstrom to the micron - a combination of different techniques is necessary to reach the macromolecular dynamics via behavior laws.
    Firstly, we validated the use of infrared spectroscopy (FTIR), differential scanning calorimetry (DSC) and thermostimulated Currents (TSC) techniques to analyze human cutaneous biopsies. These techniques were proven to be suitable to scan the dermis over a wide range of scales and to provide relevant information on the main macromolecules of the dermis and the different water structures.
    Then we studied the evolution of thermal, dielectric and vibrational signatures of the dermis during chronological aging and photo-aging via a clinical study involving subjects from two age groups (20-30 years and over 60 years). Markers of intrinsic and extrinsic aging have been extracted from this work.
    The evolution of these signatures was also studied in an ex vivo study on the effects of UV exposure of dermis, leading to a better understanding of the different mechanisms involved in chronological aging and photo-aging.
